I have the following example:

So if was to remove the content for the left div and jquery would then add a class to the right div.

How can this be done?

You need to add your code to any event listener that changes the content of the of the left div. If it's a button, your code may look like this

var DivContent = "THIS TEXT GOES IN LEFT DIV";

$('#ReplaceDivContentButton').click(function(){
    $('#left').html(DivContent);
    if ($("#left").html().length > 0) {
         $('#right').addClass('test');
    }  
 });
