Your question is very basic, which is why you're getting the snide remarks. On behalf of Stack Overflow, sorry about those.
Basically, you need to ensure that you avoid SQL Injection (very important) and that you do any validation you need to do on the input to make sure it's in the right format, it has legal values, etc.
For example, if variable1 is supposed to be someone's email address, validate it so that it actually is someone's email address and if it's not, show the user a sensible error message. Do this for any variable you accept from user input.
Using PDO is best for connecting to and using the database from PHP, but it still won't automatically mean you avoid SQL injection. To do that, you have to make use either of query parameterisation or (less secure) elaborately make sure you escape every variable before assembling it into an SQL string.
Look at PDO, and look at the two other links in the previous paragraph, and see if that gets you any closer to where you want to be.
