I would like to write a CodeIgniter library that has a command-line interface. CodeIgniter supports CLI: http://ellislab.com/codeigniter/user-guide/general/cli.html

However, I'm wondering, is there a way to define a route/controller from within a third-party library?

Only in some weird hack-ish way. Libraries are not where you want to define application processes such as routes and controllers. Your route should direct the app to a controller, which may use a library to perform various common tasks.

There are different options for creating dynamic URLs in CI apps -- maybe if you better explained the specific scenario you're trying to solve, a better-suited answer could be provided.
