I know this is a very general question and is depending on a lot of things. But I have an application that performance is varying very much from one request to another. The whole solution is something like this:

Web Application (ASP.NET) - Web Service (WCF) - Application Server - SQL Server.

The web application calls the webservice which in turn calls a COM Server that eventually goes to the database if needed. All of these components is on different servers and I'm not sure how to find the bottleneck. As I mention earlier it sometimes uses 50 seconds to load one page, while some request later or sooner the time usage is 1-3 seconds for the same page. If the code is bad written or slow it would always be, on every request, right?

On the servers, which is virtual, we have 4 GB of RAM and about 100GB of disk, some are 64-bits and some are 32-bits. Some are Win Server 2012 and some are 2008. Im looking for some general advice and best practices for setting up an application for about 100 users.

Is it OK to have an virtual environment for this kind of apps? Is 4 GB enough? What about CPU? Is there any readings on this topic I could read?

Without knowing more about your application, I can only make guesses, but it sounds like you're hitting startup costs. Depending upon how heavyweight your ASP.NET application's startup code is, it can take anywhere from a couple of seconds to a full minute for it to be able to respond to the first request, after which the worker process has been "warmed up" and remains in memory. If you're having one ASP.NET app call another, you're hitting that startup issue twice. Newer versions of IIS (7.5+) have a setting (Start Automatically) that allows you to indicate that an AppPool should be started before any requests come in, which may help here.

Thank you for your time, Andy. The performance-drop happens in the middle of a users session, there is no pattern, but it could be on a random request. I'm looking for something like: -"On a Windows 2012 64-bit server you should have at least 8 GB of RAM." Is that nothing to consider? –  Holy Crap Oct 14 '13 at 12:53

Not really. Assuming that this is the only app running on the Web and AppServer-tiers, as long as you're above a gig or so, you're not going to hit any weird slowdowns (which tend to come from things like paging to/from disk, which happens in low-physical-memory configs). Running on a VM shouldn't have this kind of dramatic impact (unless the host machine itself is a highly-utilized box). –  Andy Hopper Oct 14 '13 at 12:56
