I have a mobile application and it uses a custom REST wrapper over WordPress CMS as its backend.

WordPress is GPLv2 licensed.

If for some reason I need to modify WordPress to meet some requirement, will I be required to open source the rest of my code as well (the REST API and the mobile app)?

You need to go to the GPL2 FAQ page here and see exactly in what situation you are in and determin what obligations you have regarding the open sourcing.
