It's a vague question. But here is some advice for guidlines to go by.
Assuming you know all the technical needs, the UI should be based on the user's use of the system. Sit with the user and ask them how they do their tasks currently. Work with them to figure out how your application can fit in with the way they work and their thought process and work flow.
Once you have that, you can start to figure out how the UI would be developed. An example is if you have certain functions that they will use all the time, make it more prominant and easier to access. Things that they won't use as often can be "hidden" or made less prominent.
Another thing to do is, if possible, after you have released the application, sit with them again and get feedback on how it could be improved to work with their work flow. Since they have been using it for a while, they can provide very valuable and relevant feedback.
