Well, what is FileHelpers here, and what is loading it? For example, is this part of your config file? If so: where? What does it want? And what trust level is your site at? Give us clues... –  Marc Gravell Jun 12 '11 at 9:02

The next time you post a question, please give much more information about the environment you're using and about what you're trying to achieve. When does the error occurr ("load the site" can mean anything)? Most likely you're using IIS; you didn't mention it. What programming languages and frameworks are you using? Have a look at IIS7 Failed to grant minimum permission requests. It describes your problem and the most likely solution.
