How to delete previous marker2, after update a new marker2?

Before you add a new layer with new marker you should remove layer with old marker. So, before

marker2 = new OpenLayers.Layer.Markers( "Car" );    
map.addLayer(marker2);

you should call

map.removeLayer(map.getLayersByName('Marker')[0]);

That's quite an overkill though. You should instead use only one Marker layer and add/remove markers from it using methods addMarker and removeMarker
