Are there any built-in way to store Zend ACL into a backend storage like mysql? I'd like to manage the roles/users from the admin so the administrator can apply them on the fly. Is this a good/bad practice? If bad, what are the disadvantages?

I think it's a good practice to have this kind of dynamic management. Anyway this is a good video tutorial from which you can learn how to do it:
