What is the best approach to get the # of concurrent users of an win-based application which uses a Database as its data store?

You will need to have some kind of session associated with each user and count these. This is normally achieved with a login system. If you do not have this, adding some kind of LastActivity record and querying this will give a relatively accurate count.

You will need to think about how and when this updated. It could be via an automatic timer based approach (sent from the client) or updated when something is requested from the Data Store.
