readlines() returns a list containing all the lines in the file. While it's obviously more verbose than necessary (and will use a little more memory), there's nothing "wrong" about using readlines to explicitly get the list of lines to iterate over.

Why would you compile it for every call? The whole point of compiling it is to reuse it. Compiling the regex object once cuts the regex runtime in half on my system (it's still almost 3 times as slow as the string methods) (EDIT: Misread your compiling explanation... interesting that we got different results though)

A compiled python regex object inherits all the regex methods. So, you could simplify the substitution to: MULT_SPACES.sub(' ', 'this is a string')

It doesn't compile anything, it just lets you call an external executable. If you want to compile it, you'll have to call a compiler of some sort. The example I gave would call /usr/bin/foo. If you wanted it to call /usr/bin/foo.py, you'd need to put that in there.
