Worked with C, C# and transact SQL when employed. Done a few paid gigs as a web developer too. As a hobbyist I use C and C#. Also pretty good with Python, assembly (fasm) and C++.

Java is tricky. Applications normally uses graphics toolkits like swing rather than standard windows controls. IUIAutomation is a newer method of automating user interfaces that will work for some applications, so I'd look there. Other than that, your question is far too vague to provide any useful help.
