If you are in C:\Program Files\Java\jre6\bin directory, calling bin\debug.keystore looks for a keystore file in the directory C:\Program Files\Java\jre6\bin\bin\debug.keystore which does not exist. Leave the bin\ out.

Try putting quotes around the paths in case the spaces in the directory names is confusing things. Or try copying your keystore file into the java bin directory and calling keystool from that directory, not using paths at all.

If you do that, it's going to be looking for the keystore in C:\Program Files\Java\jdk1.6.0_21\bin\.android\debug.keystore which is almost certainly not the correct path. You need to give it the correct full path to your keystore.
